ABSTRACT

The Brazilian Cerrado biome consists of a great variety of endemic species with several bioactive compounds, and Anadenanthera peregrina (L.) Speg is a promising species. In this study, we aimed to perform phytochemical characterization and evaluate the antioxidant and antibacterial activities against Staphylococcus aureus and Escherichia coli of the hydroethanolic extract of A. peregrina stem bark. The barks were collected in the Botanical Garden of Goiânia, Brazil. The hydroethanolic extract was obtained by percolation and subjected to physicochemical screening, total phenolic content estimation, high-performance liquid chromatography (HPLC) fingerprinting, and antioxidant (IC50 values were calculated for the 2,2-diphenyl-1-picrylhydrazyl assay - DPPH) and antibacterial activity determination. The pH of the extract was 5.21 and density was 0.956 g/cm3. The phytochemical screening indicated the presence of cardiac glycosides, organic acids, reducing sugars, hemolytic saponins, phenols, coumarins, condensed tannins, flavonoids, catechins, depsides, and depsidones derived from benzoquinones. The extract showed intense hemolytic activity. The total phenolic content was 6.40 g GAE 100 g-1. The HPLC fingerprinting analysis revealed the presence of gallic acid, catechin, and epicatechin. We confirmed the antioxidant activity of the extract. Furthermore, the extract did not inhibit the growth of E. coli colonies at any volume tested, but there were halos around S. aureus colonies at all three volumes tested. These results contribute to a better understanding of the chemical composition of A. peregrina stem bark and further support the medicinal applications of this species.

ABSTRACT

Objetivou-se avaliar resposta inflamatória e concentrações de proteínas em líquido peritonealde asininas submetidas à ovariectomia por laparotomia e laparoscopia. Foram utilizadas 18 fêmeas adultas, pesando em média 100kg, as quais foram divididas em dois grupos, de nove animais cada: o grupo I, em que foram ovariectomizadas por laparotomia, e o grupo II por laparoscopia. Após as cirurgias, em ambos os grupos foram avaliadas as seguintes variáveis no líquido peritoneal: pH, densidade, número de hemácias, leucócitos e proteínas de fase aguda (fibrinogênio, haptoglobina, albumina, antitripsina, alfa-1 glicoproteína ácida, ceruloplasmina, transferrina, imunoglobulinas A e G). Todas as variáveis foram analisadas antes das cirurgias, 12, 24, 48, 72 horas, oito e 16 dias após os procedimentos cirúrgicos. O número de hemácias e leucócitos aumentou 24 horas depois das cirurgias nos dois grupos. Haptoglobina, alfa-1 glicoproteína ácida,albumina, transferrina, gamaglobulinas A, G e proteínas totais aumentaram nos dois grupos. As concentrações de ceruloplasmina não variaram enquanto a antitripsina não foi detectada no líquido peritoneal de asininos.Foi identificada uma proteína com23.000kD, que não tem denominação nem descrição no líquido peritoneal de asininos.(AU)


The aim of this study was to evaluate the inflammatory response and protein concentrations in the peritoneal fluid of donkeys submitted to laparotomy and laparoscopic ovariectomy. Eighteen adult females, weighing on average 100kg, were divided into two groups, of 09 animals each, where in group I, they were ovariectomized by laparotomy and group II by laparoscopy. Following the surgeries, the following variables were evaluated in the peritoneal fluid: pH, density, number of red cells, leukocytes and acute phase proteins (fibrinogen, haptoglobin, albumin, antitrypsin, alpha-1 acid glycoprotein, ceruloplasmin, transferrin, Immunoglobulins A and G). All variables were analyzed before the surgeries, 12, 24, 48, 72 hours, 8 and 16 days after the surgical procedures. The number of red cells and leukocytes increased 24 hours after the surgeries in both groups. Haptoglobin, alpha-1 acid glycoprotein, albumin, transferrin, gammaglobulins A and G, and total proteins increased in both groups. Concentrations of ceruloplasmin did not change while antitrypsin was not detected in the asinine peritoneal fluid. A protein of 23,000kD, which has no name or description in the peritoneal liquid of donkeys, has been identified.(AU)

ABSTRACT

Os frutanos do tipo inulina são oligossacarídeos que favorecem a multiplicação de determinados gêneros bacterianos no intestino, promovendo um efeito prebiótico. Este trabalho avaliou o efeito da inulina extraída de raízes de yacon (Smallanthus sonchifolius) sobre a colonização intestinal de frangos de corte experimentalmente infectados por Salmonella Enteritidis. Sessenta frangos de corte com um dia de idade foram divididos em três grupos de tratamento, com duas repetições, criados até 21 dias. As aves do grupo yacon receberam 100mg de inulina/dia, via oral, por três dias consecutivos. No sétimo dia de vida, as aves tratadas e o controle positivo foram desafiados pela via oral com uma cultura de S. Enteritidis. Não foram observadas diferenças de desempenho zootécnico entre os grupos. O índice de infectividade das aves suplementadas com yacon foi menor até o sexto dia após o desafio, mas, ao término do experimento, foi superior ao controle positivo. Os dados deste trabalho demonstram que o uso da inulina nos três primeiros dias de vida promoveu uma redução da colonização intestinal dos frangos por Salmonella Enteritidis na primeira semana após o desafio. Novos estudos são necessários para determinar a dose e o tempo de tratamento ideal para um efeito protetor de maior duração.(AU)


The fructan inulin-type oligosaccharides favor the multiplication of some bacterial genera in the intestine, promoting a prebiotic effect. This study evaluated the effect of inulin extracted from yacon roots (Smallanthus sonchifolius) on intestinal colonization of broilers experimentally infected with Salmonella Enteritidis. Sixty-one day old chicks were grouped into three treatments, with two replicates, and reared until 21 days. Birds in the yacon group received 100mg of inulin/day orally for three consecutive days. On the seventh day of life the treated birds and the positive control were challenged orally with a culture of S. Enteritidis. There were no differences between groups in live performance. The infectivity index of the chicks supplemented with yacon was lower until the sixth day after the challenge, but at the end of the experiment it was higher than the positive control. Data from this study show that the use of inulin during the first 3 days of life caused a reduction of intestinal colonization of chickens by Salmonella Enteritidis in the first week after challenge. Further studies are needed to determine the dose and the ideal time of treatment necessary for a longer protective effect.(AU)

ABSTRACT

It has been demonstrated that resistance exercise improves cognitive functions in humans. Thus, an animal model that mimics this phenomenon can be an important tool for studying the underlying neurophysiological mechanisms. Here, we tested if an animal model for resistance exercise was able to improve the performance in a hippocampus-dependent memory task. In addition, we also evaluated the level of insulin-like growth factor 1/insulin growth factor receptor (IGF-1/IGF-1R), which plays pleiotropic roles in the nervous system. Adult male Wistar rats were divided into three groups (N = 10 for each group): control, SHAM, and resistance exercise (RES). The RES group was submitted to 8 weeks of progressive resistance exercise in a vertical ladder apparatus, while the SHAM group was left in the same apparatus without exercising. Analysis of a cross-sectional area of the flexor digitorum longus muscle indicated that this training period was sufficient to cause muscle fiber hypertrophy. In a step-through passive avoidance task (PA), the RES group presented a longer latency than the other groups on the test day. We also observed an increase of 43 and 94% for systemic and hippocampal IGF-1 concentration, respectively, in the RES group compared to the others. A positive correlation was established between PA performance and systemic IGF-1 (r = 0.46, P < 0.05). Taken together, our data indicate that resistance exercise improves the hippocampus-dependent memory task with a concomitant increase of IGF-1 level in the rat model. This model can be further explored to better understand the effects of resistance exercise on brain functions.

ABSTRACT

The Delphi technique has been used since the 1950s to collect the opinions of experts; to gauge their indications, and in some instances, to develop a consensus. This systematic collection and aggregation of informed judgments from a group of experts on specific questions or issues is a highly efficient and cost-effective means to establish guidelines and policies, when compared to other strategies, such as committee meetings or personal interviews. OBJECTIVE: Examine the content validation process of the proposed criteria of the American Society of Health System Pharmacists (ASHP) for amikacin use in hospital settings. MATERIAL AND METHODS: The Delphi technique was applied using the proposed ASHP criteria questionnaire containing 102 specific questions related to the nosocomial use of amikacin by individual patients. The questionnaire contained six groups of questions: 1) Identification and basic demographic data, 2) Relevant data for the use of amikacin, 3) Justification of its usage, 4) Critical parameters of amikacin use, 5) Complications, 6) Measurement of results. Eight hospital specialist medical doctors were selected, including five in the area of infectious diseases, one surgeon, one nephrologist and one in critical care medicine. The questionnaire was e-mailed to the doctors and they were asked for their opinion about the appropriateness of the questions. They were to say whether the general concept seemed totally or partially adequate to the proposed process, what grade (0 to 10) they would give to each section, and if there were any perceived deficiencies, they could add, omit or modify individual questions. A second questionnaire containing the questions for which there had been no consensus based on the answers to the previous one was re-sent to the participants for consolidation. RESULTS: Feedback revealed an agreement of 75 percent concerning the utility and appropriateness of sections 1 and 2. The section about the justification of amikacin usage was agreed on by 50 percent. There was a total agreement of 62 percent for the critical parameters of amikacin use, and a partial agreement of 37 percent. The complication of usage of the questionnaire was agreed upon by 50 percent of the participants, and positive measurement of the results was totally agreed on by 62 percent, and partially by 37 percent. The overall score for the questionnaire was 8.77 ± 0.25. CONCLUSION: The usage criteria for amikacin recommended by ASHP were validated by the Delphi technique for utilization in Brazilian hospital settings. The Delphi technique applied to validate a questionnaire instrument for monitoring the correct use of a specific strategic antibiotic indicated for the treatment and prophylaxis of serious antibiotic-resistant Gram-negative bacteria, proved to be a reliable and simple tool for designing guidelines and a consensus document for hospital use of antibiotics.

ABSTRACT

This study was undertaken to verify the effect of a daily intake of a new fermented soy milk produced with Enterococcus faecium and Lactobacillus jugurti on the serum lipid levels in normocholesterolemic middle-aged men. The study was randomized, double-blind and placebo-controlled and was performed for a period of 6 weeks. Forty-four normocholesterolemic healthy, male volunteers, aged 40-55 years old were randomly separated in two groups: The F-group received 200 ml of the fermented product daily and the P-group received 200 ml of placebo (chemically fermented). The blood samples were drawn initially and after 3 and 6 weeks and serum values for total cholesterol, HDL-cholesterol and triglyceride were determined. The LDL-cholesterol value was estimated. No significant changes in the fermented group (F) were observed for total cholesterol, LDL-cholesterol or triglyceride levels, while the HDL-cholesterol level was significantly higher (p < or = 0.05) after 6 weeks. The total cholesterol and LDL-cholesterol levels were significantly higher (p < or = 0.05) in the placebo group (P), but no changes were found for the HDL-cholesterol and triglyceride levels during the experimental period. In conclusion, the intake of 200 ml/day of the fermented soy milk, produced with E. faecium and L. jugurti, for 6 weeks, did not affect the serum total cholesterol and LDL-cholesterol, and led an increase of 10 per cent in the HDL-cholesterol level.

ABSTRACT

A aterosclerose pode ser considerada uma doença na circulaçäo coronariana. O presente trabalho estuda a açÄo da quercetina, morina, ácido nicotínico de maneira isolada e em associaçÄo envolvendo quercetina + ácido nicotínico e morina + ácido nicotínico no metabolismo lipídico. Foram dosados colesterol, colesterol-HDL e triacilglicerol após administraçäo de duas doses dos compostos morina, quercetina, morina + ácido nicotínico e quercetina + ácido nicotínico sendo a primeira imediatamente após a administraçäo do triton e a segunda dose, vinte horas depois. Decorridos quarenta e três horas após a administraçäo do triton o sangue foi analisado. Os resultados mostraram que morina + ácido nicotínico e quercetina + ácido nicotínico apresentaram os melhores resultados para colesterol (-83,77 porcento e 74,42 porcento). Para colesterol-HDL os melhores resultados foram com morina e com associaçäo quercetina + ácido nicotínico (+17,99 porcento e +21,96 porcento). Morina + ácido nicotínico mostraram os melhores níveis para triacilgliceróis (-83,77 porcento)

ABSTRACT

A single electroconvulsive shock (ECS) or a sham ECS was administered to male 3-4-month-old Wistar rats 1,2, and 4 h before training in an inhibitory avoidance test and in cued classical fear conditioning (measured by means of freezing time in a new environment). ECS impaired inhibitory avoidance at all times and, at 1 or 2 h before training, reduced freezing time before and after re-presentation of the ECS. These results are interpreted as a transient conditioned simulus (CS)-induced anxiolytic or analgesic effect lasting about 2 h after a single treatment, in addition to the known amnesic effect of the stimulus. This suggests that the effect of anterograde learning impairement is demonstrated unequivocally only when the analgesic/anxiolytic effect is over (about 4 h after ECS administration) and that this impairment of learning is selective, affecting inhibitory avoidance but not classical fear conditioning to a discrete stimulus.

ABSTRACT

A partir de um surto de Leishmaniose Tegumentar (LT) humana no Triângulo Mineiro e Alto Paranaíba, propusemo-nos investigar a infecçäo natural em cäes em áreas rural e urbana do Município de Uberlândia - Minas Gerais. Os cäes foram submetidos a exames clínico, hemoparasitológico e sorológico (reaçäo de imunofluorescência indireta). Foram examinados 429 cäes sendo 100 da área rural e 329 da área urbana. A RIFI demonstrou que 1 por cento dos animais da zona rural e 10,6 por cento dos animais da área urbana foram reagentes para Leishmania. Este achado parece indicar que a infecçäo canina na regiäo de estudo é ativa e que o cäo poderá representar um importante papel como reservatório da infecçäo, podendo participar na disseminaçäo e formaçäo de novos focos de endemia também em área urbana

ABSTRACT

1. The effect of acute ethanol on memory was studied in an eight-arm radial maze by interposing a 15-s or 1-h delay between the rat's fourth and fifth arma choices. 2. Ethanol (1.0g/Kg) was injected intraperitoneally 5 min prior to the firsrt set of 4-arm choices, therefore being presrnt since the acquisition of the trial-unique event. 3. The results showed 1) a decrease in choice accuracu only in the final 4 arm choices after the 1-h delay, and 2) that errors consisted of re-entries into arms chosen before the delay was imposed. The data further support the contention that ethanol impairs retention of working memory

ABSTRACT

Os autores analisam os acidentes de trabalho ocorridos em um hospital geral no periodo compreendido entre janeiro e dezembro de 1979 e a sua distribuicao entre os funcionarios, objetivando confirmar resultados anteriores e avaliar uma possivel existencia de funcionarios pertencentes a outras categorias profissionais que estariam expostos a riscos. Concluem afirmando que, tambem, e mais elevada a incidencia de acidentes em funcionarios enquadrados em menores faixas salariais, como cozinheiros, seventes de nutricao e de lavanderia e atendentes de enfermagem. Acrescentam, no entanto, a essa relacao outros grupos constituidos por marcineiros - atividade de maior risco - e mecanicos.Destacam a importancia do numero de acidentes com escriturarios e mensageiros, embora esses acidentes se situem entre as categorias de menor risco. Finalmente, propoem que uma estrategia prevencionista em um hospital geral deva levar em consideracao estes achados

ABSTRACT

Dois grupos de pacientes foram submetidos a operacao cesariana por indicacoes diversas na Clinica Obstetrica do Hospital Barao de Lucena (HBL), em Recife, ambos com rotura das membranas cujo tempo de latencia era de, no maximo, 12 horas por ocasiao da laparo-histerotomia. Num dos grupos, foi utilizado o cloranfenicol profilaticamente e, no outro, nao se fez uso de antibiotico. A morbidade materna foi avaliada atraves dos parametros febre e abcesso de parede, nao se mostrando util o antibiotico na profilaxia da infeccao. O tempo de permanencia hospitalar foi praticamente identico nos dois grupos investigados
